In this thesis (R)-3-hydroxycarboxylic acids (HA) from bacterial polyhydroxyalkanoates (PHA) were investigated, regarding their biotechnological production as well as intracellular processes of their formation and degradation. An innovative bioprocess to provide HA as fine chemicals for further applications is presented. The method is straightforward, easy to handle and results in high yields (introduced in chapters 2-4). Following the observations of this study, attempts to scale up the process seemed possible and potential applications of HA remain topics of research. The underlying mechanisms of PHA degradation were also addressed, including issues of physiological advantages of HA excretion for cells (chapter 5) and characterization of enzymes having HA as substrates (chapters 6 and 7). Even though some aspects of PHA metabolism were clarified for P. putida GPo1, many details about intracellular PHA degradation remain obscure and require further experiments and discussion. Knowledge about their molecular context in the bacterial cell is crucial for efficient PHA and HA production.

Lumpy skin disease (LSD) is caused by a virus in the genus Capripoxvirus of the family Poxviridae. The virus has been reported to be excreted in the semen of experimental infected nonvaccinated bulls. Nevertheless, vaccination has been the most widely used method to reduce and prevent the spread of the disease. This work was done to determine the efficacy of lumpy skin disease vaccination in preventing the excretion of lumpy skin disease virus(LSDV) in semen of experimentally infected vaccinated bulls. It also determined further the effect of vaccination and experimental infection on semen quality. Six serologically negative bulls were vaccinated with an attenuated Neethling strain of LSD vaccine. These bulls were then experimentally infected by intravenous injection with a virulent field strain of LSDV. Six unvaccinated bulls were similarly infected to act as controlsThe current data supports the inclusion of a LSD vaccination programme using this vaccine as a biosecurity measure with regards to the movement of semen from LSD infected areas.

In this study it was assessed the effects of dietary L-Carnitine on lipid metabolism in African catfish (Clarias gariepinus, Burchell). The working hypotheses of this book were the following. Firstly, growth and fatty acid concentrations in fish are positively related to dietary carnitine levels. Carnitine supplementation is expected to increase fatty acid oxidation and as a result lean tissue content will increase. Secondly, one may expect that nutritional conditions that result in decreased carnitine synthesis will enhance the effect of dietary carnitine. Thirdly, the increased lipid oxidation, resulting from the extra dietary carnitine, may favour the protein-sparing effect. Finally, extra dietary carnitine may improve the energy status of the working muscle, thus protecting it against sudden energy depletion, often experienced by fish exposed to prolonged exercise. In conclusion, dietary carnitine supplementation altered the non-protein energy metabolism and decreased postprandial ammonia excretion. Factors that modulate the action of dietary carnitine is discussed in this book.
